New Study Validates MariGuard® Lutein and Meso Zeaxanthin 1:2 Ratio as the Scientific Standard for Ocular Health

A just-released study shows that a 1:2 ratio of MariGuard® Lutein to Meso-zeaxanthin, matching the ratio naturally present in the human macula, provides significantly greater protection against oxidative damage to retinal cells than any other tested combination.

The research team, composed of specialists from INNOBIO and a collaborating university, published its findings in the American Journal of Biomedical Science & Research. The study, titled "The Effects of Lutein: Meso-Zeaxanthin =1:2 on Oxidative Stress-Induced Retinal Damage via Up-Regulation of MTHFD1L" shows the first evidence of the synergistic efficacy of this specific 1:2 combination.
Notably, this 1:2 ratio mirrors the natural pigment profile found in the human macula. As such, the study marks a pivotal shift – moving the industry away from ad hoc blending and toward a scientifically validated formulation standard for clinical applications.

Efficacious Cellular Protection Confirmed

The investigation compared multiple lutein to meso-zeaxanthin ratios – 5:1, 2:1, 1:1, and 1:2, along with blends containing other zeaxanthin isomers (3R-3'R-zeaxanthin), using both ARPE-19 retinal cells and mouse models.

The 1:2 lutein:meso-zeaxanthin ratio outperformed all other formulations across multiple endpoints:

  • Increased cell viability under oxidative stress
  • Reduced MDA (malondialdehyde) levels, a key marker of lipid peroxidation
  • Enhanced activity of antioxidant enzymes (T-SOD, GSH-Px)
  • Effective scavenging of ROS (reactive oxygen species) in retinal pigment epithelial cells

In mouse models, the 1:2 formulation demonstrated significant tissue-level benefits. Histological analysis showed that it reversed atrophy in both the outer and inner nuclear layers of retina and preserved the normal photoreceptor architecture – effects not observed with other ratios.

Discovery of a new molecular pathway

Beyond confirming efficacy, the research unveiled the mechanistic basis: the 1:2 ratio activates both the canonical Nrf2/HO-1 antioxidant pathway and the novel target MTHFD1L. This dual action intercepts oxidative damage at the cellular level, establishing a new theoretical framework for precision ocular nutrition.

"Meso-zeaxanthin is the dominant pigment in the fovea, where the physiological lutein to meso-zeaxanthin ratio is approximately 1:2," said an INNOBIO representative."Our study proves that replicating this exact ratio delivers clinical benefits unattainable with any other combination."

Bridging science and commercial application

This breakthrough is enabled by INNOBIO's proprietary caroblast™ microencapsulation technology, designed to overcome carotenoids’inherent challenges—poor stability, low solubility, and suboptimal bioavailability. Independent lab data show that microencapsulation boosts cellular uptake of lutein and meso-zeaxanthin by 3.8-fold and 3.6-fold, respectively, ensuring that the 1:2 synergistic ratio is effectively delivered in finished products.

MariGuard® is available in multiple formats-liquid suspensions, microencapsulated powders, and beadlets, including a ready-to-drink microencapsulated powder specifically designed for the liquid beverage sector. These formats are compatible with softgels, hard capsules, tablets, gummies, liquid beverages, and solid drinks, offering manufacturers broad formulation flexibility.

Industry Implications

The study's findings arrive at a critical juncture, as consumers increasingly demand science-backed eye health solutions. This research establishes an evidence-based benchmark for lutein and meso-zeaxanthin products, effectively bridging formulation science with physiological research.

“Innovation in this industry isn't just about sourcing ingredients – it's about understanding the complex clinical science behind them,” said a company spokesperson. “We are committed to both foundational and future research, ensuring our partners have all they need to develop next-generation eye health products.”
